I'm developing what's called a mutant library, based on one of the RNA sequences that survived my SELEX experiment. Preliminary reporter experiments with this RNA, 36 (yet to be given a name), may regulate gene expression in response to ribosomal protein S15 (from G. kaustophilus species). I mutated the RNA sequence and now need to screen my mutants to find which sequences actually DO regulate gene expression using a GFP reporter.
